Key Facts

1AMVUTTRA net revenue reached about 1,011.762 million dollars in the second quarter of 2026, up 106% year over year.
2Higher demand lifted U.S. TTR revenue by 129 million dollars quarter over quarter, while the inventory impact was about 20 million dollars.
3Alnylam cut its 2026 TTR product revenue forecast to 4,200–4,500 million dollars.
4The highest dose group in the Phase 1b study showed a mean 60–64% reduction in mutant huntingtin protein after 3 months.

Alnylam Pharmaceuticals said net revenue from AMVUTTRA reached 1,011.762 million dollars in the second quarter of 2026, up 106% year over year from 491.953 million dollars in the same period of 2025. The results were reported in the company’s second-quarter financial statement, while on October 23 it announced preliminary results from a Phase 1b study of its experimental drug brezosiran for Huntington’s disease. The figures reflect strong growth in commercial treatment sales alongside progress in an early-stage research program. In July, however, the company cut its annual revenue forecast for TTR products, the category that includes AMVUTTRA. Alnylam said the adjustment was tied to moderating U.S. demand from patients starting treatment in the second line, after the early launch benefited from patients who had been waiting for a new treatment option.

Alnylam’s total revenue rose to 1,290.948 million dollars in the second quarter from 773.689 million dollars a year earlier, an increase of 67%. Net product revenue climbed 74% to 1,172.109 million dollars, from 672.212 million dollars in the second quarter of 2025. The company also reported U.S. GAAP operating income of 231.441 million dollars, compared with an operating loss of 16.199 million dollars a year earlier. The company said AMVUTTRA growth was driven mainly by increased demand among U.S. patients with transthyretin amyloid cardiomyopathy. U.S. TTR product revenue rose 106 million dollars from the first quarter of 2026, with higher demand contributing 129 million dollars, partly offset by an inventory impact of about 20 million dollars and a slight reduction in net price.

Internationally, TTR product revenue increased by 14 million dollars from the first quarter, supported mainly by demand for treatments for transthyretin amyloid polyneuropathy and cardiomyopathy. The company expanded AMVUTTRA’s rollout with a launch in Spain and entered a commercial partnership with BeOne Medicines to distribute it in mainland China and Macau, subject to marketing approval. Alnylam said the first quarter of 2026 was the first quarter in which product revenue exceeded 1 billion dollars, followed by TTR revenue exceeding 1 billion dollars in the second quarter. But the company lowered its 2026 TTR product revenue forecast range from 4,400–4,700 million dollars to 4,200–4,500 million dollars. It also cut its forecast for total net product revenue from 4,900–5,300 million dollars to 4,700–5,100 million dollars, citing a return of AMVUTTRA demand growth in the U.S. second-line treatment setting to what it described as a normal level.

For brezosiran, data from the highest dose group evaluated so far showed a mean reduction of about 60–64% from baseline in mutant huntingtin protein in cerebrospinal fluid after 3 months, while the largest reduction in an individual participant was 83%. The preliminary data covered 48 participants across 5 groups that received a single dose, while the company was enrolling 12 additional participants in a sixth dose group and had not yet evaluated their results. The study followed some participants for up to 12 months, and the company said it had observed no dose-dependent increases in cerebrospinal fluid inflammatory markers, neurofilament light chain or brain ventricular volume. Alnylam presented the results as preliminary safety, tolerability and biomarker data, not as evidence of established therapeutic efficacy or that the treatment slows the disease. The company plans to begin a placebo-controlled pivotal study in 2027, subject to regulatory alignment.

For upcoming events, Alnylam scheduled a discussion of its Huntington’s program at the RNAi Roundtable on October 26, 2026, at 10:00 a.m. Eastern time. The company also scheduled a call to discuss its third-quarter 2026 results for October 29, after announcing the results discussion date on October 15. ALNY shares closed at 224.03 dollars on October 23, down from 227.28 dollars in the previous session. The company also expects to announce data in the second half of 2026 from Phase 1 and Phase 2 studies of ALN-6400, and Phase 1 results for ALN-2232. The planned start of the pivotal brezosiran study in 2027 remains subject to regulatory alignment.
