Key Facts

1The FDA supported a single two-year registration trial plan for WVE-006, with an interim analysis after one year.
2Day 85 measurements showed a 9.2% reduction in visceral fat and a 4.0% reduction in total fat after adjustment for placebo.
3Wave had $490.6 million in cash and marketable securities on June 30, 2026.
4WVE shares closed at $3.35 on October 22, 2026.

Wave Life Sciences said on October 1 that, following a meeting with the U.S. Food and Drug Administration, the agency supported the design of a single two-year registration trial for WVE-006, including an interim analysis after one year. The company said the analysis could support an application for accelerated approval based on markers associated with alpha-1 antitrypsin deficiency, while the trial is intended to support the possibility of full approval. It said the agency also supported its current mass spectrometry assay. WVE-006 is being studied in an open-label Phase 1b/2a trial that includes people with alpha-1 antitrypsin deficiency and the homozygous Pi*ZZ mutation. The plan remains a potential registration pathway, and the company has not announced approval of the treatment. Wave is scheduled to present data from the 600 mg monthly multiple-dose cohort in the fourth quarter of 2026, and said the treatment had not been associated with liver toxicity as of its announcement.

The numerical WVE-007 data cited in earlier coverage were published by the company on December 8, 2025, and are not new results as of October 23, 2026. In the Phase 1 INLIGHT study, participants were randomized 3 to 1 between treatment and placebo, and the 240 mg single-dose cohort included 32 participants. After adjustment against placebo, measurements on day 85 showed a 9.2% reduction in visceral fat and a 4.0% reduction in total fat mass. By contrast, lean mass increased 0.9% and total mass decreased 0.9%. The company said WVE-007 was generally safe and well tolerated up to a 600 mg dose, with no treatment-related withdrawals or serious or severe adverse events recorded as of the date of its statement. These results place the program at an early stage, while the company continued to follow participants after the initial measurement.

In an update issued in March 2026, Wave said six-month follow-up after a single dose showed a 14.3% reduction in visceral fat and a 5.3% reduction in total fat mass compared with placebo, alongside a 2.4% increase in lean mass. The company also compared the visceral-fat-to-muscle ratio with results from another semaglutide trial. It said the ratio fell 16.5% from baseline in INLIGHT, compared with a 12.2% decline in the BELIEVE trial after six months. It noted a difference in the groups' average body mass index, which was 32 kg/m² among INLIGHT participants and 37 kg/m² in BELIEVE. The company thus presented the follow-up results as a comparison between two studies that differed in average body mass index, alongside measurements of fat and muscle mass. This comparison is not a direct trial result combining the two treatments.

In September 2026, Wave moved to test WVE-007 with tirzepatide in a combination Phase 2a trial. The study is testing WVE-007 doses of 240 and 400 mg alongside weekly tirzepatide in people with obesity and type 2 diabetes. The company set the participants' body mass index range at 35 to 50 kg/m². It also said it aims to begin a clinical maintenance trial in the second half of 2026 to examine the use of WVE-007 to maintain weight loss after stopping incretin treatment. Regarding its financial resources, the company reported cash, cash equivalents and marketable securities of $490.6 million on June 30, 2026. It said its liquidity is expected to last through the third quarter of 2028.

WVE shares closed at $3.35 on October 22, 2026, according to EL7 data. The company scheduled its third-quarter results announcement and investor call for October 27 at 8:30 a.m. Eastern Time. It also announced that its virtual R&D day will be held on November 4, 2026. Among the upcoming clinical milestones, Wave aims to present data from the WVE-006 600 mg monthly cohort in the fourth quarter of 2026. The market is therefore watching multiple milestones across the alpha-1 antitrypsin deficiency and obesity programs, with different data and regulatory timelines for each.
