FDA Accepts Unicycive Drug Resubmission for 6-Month Review
Key Facts
Unicycive Therapeutics announced on October 21, 2026 that the U.S. Food and Drug Administration had accepted the resubmission of its application for approval of oxylanthanum carbonate. The company said in its statement that the agency classified the resubmission as Class 2, subject to a 6-month review period. That period runs from the date the application was resubmitted to the regulator. The application concerns an investigational drug intended to treat hyperphosphatemia in patients with chronic kidney disease who are on dialysis. Acceptance followed the addition of manufacturing data from an alternative supplier, after problems at the original third-party manufacturer stalled the previous review.
The company had said the latest complete response letter was based on "the same third-party manufacturing deficiencies." It added that an inspection of that manufacturer's facility did not take place during the review of the previous resubmission. By contrast, it said the FDA had raised no concerns about the clinical efficacy or safety data submitted in the application. The new package included chemistry, manufacturing and controls data from a new drug product manufacturer, according to the resubmission statement. The company thus submitted data in the current application from an alternative to the manufacturer associated with the deficiencies cited in the agency's letter.
Unicycive said the alternative manufacturer had produced the drug and completed 12-month product stability studies. The company also included in vitro bridging data between the products from the two manufacturers in the resubmission package. According to its statement, the alternative manufacturer's facility was last inspected by the FDA in March 2024. The facility received a "No Action Indicated" classification at that time, according to the inspection outcome announced by the company. Unicycive intends to retain both drug product manufacturers to provide a backup in its supply chain rather than rely on a single manufacturer.
Oxylanthanum carbonate is an investigational lanthanum-based phosphate binder the company is developing for the target population of kidney patients. Its development uses nanoparticle technology intended to reduce the number and size of pills patients take. The company says this approach may help improve adherence to treatment. The application for approval is based on 3 clinical studies, alongside preclinical and manufacturing data. The package includes a Phase 1 study conducted in healthy volunteers and a bioequivalence study. The other study examined the drug's tolerability in kidney patients on dialysis, the population targeted by the treatment.
On funding, Unicycive held $61.4 million in cash, cash equivalents and marketable securities as of June 30, 2026, according to unaudited figures announced by the company. In its resubmission update, it projected that its cash resources would fund operations into the second half of 2027. During the current review, the company said it did not know whether the FDA would conduct a pre-approval inspection of the alternative drug product manufacturer. Potential approval remains contingent on completion of the review and regulatory requirements related to manufacturing and inspection. The agency set March 25, 2027 as the target date for regulatory action on the application under the Prescription Drug User Fee Act.
